Multiplexed Bead-based High Throughput Screening
Utilizing multiplexed beads with HyperCyt dramatically increases the potential numbers of results possible per well. The assay described here is a no-wash fluorescent GTP-binding assay adapted to multiplexed, high-throughput measurements whereby multiple GTPases are simultaneously screened against the MLSCN library.
- Four micron beads with varying fluorescent intensities were analyzed on the HyperCyt platform connected to a CyAn flow cytometer.
- Beads from multiple wells are shown in this array, illustrating the ability to analyze multiplexed bead sets with HyperCyt.
- Beads were incubated with test compounds or buffer in the presence of Bodipy-Fl-GTP for 40 minutes at 4°C. The FL-1 vs time histogram illustrate the ability to view dose responses to compound blocking activity.
